Larazotide acetate is a synthetic, eight-amino-acid peptide that was developed to function as a tight junction regulator. In celiac disease, the ingestion of gluten can lead to an inflammatory response that opens the "tight junctions"—the connections between the cells of the intestinal lining. This allows more gluten fragments to pass into the gut lining, which perpetuates the inflammation.
Larazotide acetate is designed to remain in the gut and is not significantly absorbed into the bloodstream. It works by acting as a zonulin antagonist, blocking the process that causes these tight junctions to open.
- Reduced gastrointestinal symptoms: In Phase 2 trials, patients with celiac disease who were following a gluten-free diet but still experiencing symptoms reported a significant reduction in discomfort, abdominal pain, and fatigue when taking a specific low dose
- Blocked immune response: Some studies showed that the drug could reduce the gluten-induced immune reaction by preventing the rise of anti-tissue transglutaminase (tTG) antibodies, which are a marker of the disease.
- Improved intestinal permeability: While not all studies showed a significant change in measured intestinal permeability (lactulose-mannitol ratio), there was evidence that the drug could prevent an increase in permeability when patients were challenged with gluten.
Store in a cool, dry place away from direct sunlight. Reconstituted peptides should be refrigerated at 2-8°C and used within 30 days. Do not freeze reconstituted solutions.
